Abstract: Fungi are organisms with high levels of bioprospecting, however, there are limitations to their correct study, such as: difficulty of collection, identification and in vitro preservation. For this reason, this research work, has the following purpose, the implementation and evaluation of methods for the conservation of basidiomycete fungal strains (ie, Armillaria sp., Gymnopus sp., Lentinus sp., and Pleurotus sp.) and Ascomycetes (ie, Pestalotiopsis sp.) in a long term. Two conservation methods were assement, the first one is the Perlite Homolka Protocol (-80ºC) Modified and the second one the PDA Medium Protocol. On these mycelium of the different strains were inoculated and monitored after monthly reactivation during a year. The data obtained indicate a reactivation of 100% in the PDA medium for the strains Gymnopus sp., Lentinus sp., and Pestalotiopsis sp. however, the Homolka Perlite modified method (-80ºC) allowed to keep the macro and microscopic characteristics for all the strains, for this reson we suggest this method and medium as the most generalist for the evaluated species. These data indicate that the strains react differently according to the conservation method.
Description: Los hongos son organismos con elevados potenciales de bioprospección, no obstante, existen limitaciones para su correcto estudio, como, por ejemplo: dificultad de colección, identificación y la preservación in vitro. Por tal motivo, este trabajo investigativo, tuvo como finalidad, la implementación y evaluación de métodos para la conservación de cepas fúngicas Basidiomicetos (i.e Armillaria sp., Gymnopus sp., Lentinus sp., y Pleurotus sp.) y Ascomicetos (i.e. Pestalotiopsis sp.) a largo plazo. Dos métodos de conservación fueron experimentados, así está el Protocolo Homolka Perlita (-80ºC) modificado y Protocolo medio PDA, donde se inoculó micelio de las diferentes cepas y se monitoreó su reactivación mensualmente durante un año. Los datos obtenidos indican una reactivación del 100% en el medio PDA para las cepas Gymnopus sp., Lentinus sp., y Pestalotiopsis sp. No obstante, el método Homolka Perlita (-80ºC) modificado permitió reactivar y mantener iguales características macro y microscópicas para todas las cepas mostrándose como, el más generalista para las especies evaluadas. Estos datos indican que las cepas reaccionan de forma distinta según el método de conservación.
