Description: Biomining is another alternative in terms of being more economic, effective and environmentally less harmful alternative; to traditional mining. The process of biooxidation the microorganisms oxidates the minerals, then transforms the components of sulfur into sulfuric acid to liberate the metals of interest. Our objective was to obtain, isolate and evaluate the capacity of oxidation in a culture of thermophilic microorganisms, from deposit of minerals in locations of Portovelo and Baños and these oxidize sulfur more efficiently and live under hard conditions of temperature, over 60º C. We took microorganism samples of sulfur oxidation from both minerals deposit, these were compared with a strain of S. metallicus donated by the Pontíficia Universidad Católica of Valparaiso. Microorganisms were cultivated at a specific liquid design, changing the energy sources and pH. During the biooxidation we determine the concentration of sulfur, pH and total protein; samples from Baños treated with ferrous sulfate at initially pH 1.8, reach the best results: 0, 7737 g L-1d-1 of sulfate and a velocity of growth of 0, 0578 h-1 at a final pH 0.6. Later we obtain DNA and amplified a specific gene region at 16S in which we obtain bands of Domain Archaea.
Biominería se da como una alternativa más económica, eficaz y menos nociva con el ambiente; ante la minería tradicional. En el proceso de biooxidación los microorganismos oxidan los minerales, transformando los compuestos de azufre en ácido sulfúrico para liberar los metales de interés. Nuestro objetivo fue obtener, aislar y evaluar la capacidad oxidativa de un cultivo de microorganismos termófilos, provenientes de yacimientos mineros de Portovelo y Baños que oxiden azufre con mayor eficiencia y vivan a temperaturas sobre los 60º C. Se obtuvieron cultivos de los dos yacimientos, los mismos que se compararon con una cepa de S. metallicus donada por la Pontificia Universidad Católica de Valparaíso. Dichos microorganismos se cultivaron en medio líquido específico, variando fuentes de energía y pH. Durante la biooxidación se determinó concentración de sulfato, pH y proteína total, con los cuales se consideró que la muestra de Baños tratada con sulfato ferroso a pH inicial 1,8 alcanzó los mejores resultados, siendo éstos: 0,7737 g L-1d-1 de sulfato, una velocidad de crecimiento de 0,0578 h-1 y un pH final de 0,6. Posteriormente, se obtuvo ADN y se amplificó la región del gen 16S en la que se obtuvieron bandas para el Domino Archaea.
