Description: The cultivation of Streptomyces through maceration in AcOEt and Amberlite® XAD-7HP obtained with a performance of 0.05%, which was fractionated by chromatography on Sephadex LH-20 using two different mixtures of solvents. Kinetic parameters were evaluated: volumetric speed and specific growth (μmax) reaches its maximum value at 48 hours, while 96 hours get maximum metabolic production with a slow generation time of 51 hours. In the toxicity test with H2O2 wasn’t revealed significant differences in biomass production of the culture, but there was a drastic decrease in the production of secondary metabolites. The total extract showed important activity against E. faecalis (CMI 0,78 μg/mL), as well the F2 fraction (CMI 0,19 μg/mL). It established an easy method to quantitatively detect metabolites present in the extracts and fractions of interest of Streptomyces by HPLC-DAD, the efficiency of this method allowed to obtain defined peaks at different retention times.
Del cultivo de Streptomyces, mediante maceración en AcOEt y extracción con Amberlite® XAD-7HP se obtuvo un extracto con rendimiento del 0,05%, el cual se fraccionó por cromatografía en Sephadex LH-20 empleando dos mezclas de disolventes diferentes. Se evaluaron parámetros cinéticos: velocidad volumétrica y específica de crecimiento (μmax) alcanzan su máximo valor a las 48 horas, mientras que a las 96 horas consigue la máxima producción metabólica con un tiempo de generación lento de 51 horas. En la prueba de toxicidad con H2O2 no se revelaron diferencias significativas en la producción de biomasa del cultivo, pero si una reducción drástica en la producción de metabolitos secundarios. El extracto total presentó una fuerte actividad frente a E. faecalis (CMI 0,78 µg/mL), así como la fracción F2 (CMI 0,19 µg/mL). Se estableció un método fácil para detectar cuantitativamente los metabolitos presentes en el extracto total y las fracciones de interés de Streptomyces mediante HPLC-DAD, la eficiencia del método permitió obtener picos definidos a diferentes tiempos de retención.
