Abstract: It was performed the isolation and characterization of secondary metabolites present in Gaiadendron punctatum, used by Saraguro culture in the province of Loja (Ecuador), in the healing of various diseases, especially related to the respiratory tract. The research started with the collection of the species in the Saraguro canton. The flowery botanical specimens were dried, weighed and leaves and flowers were classified. The extract was obtained by succesive maceration with solvents or mixture of different polarity: Leaves (hexane, ethyl acetate, methanol, methanol:water (7:3) and ethanol) and flowers (hexane, ethyl acetate and methanol). The richest extracts by TLC analysis (ethyl acetate and ethanol in leaves and hexane and methanol in flowers) were fractionated by column chromatography. The resulting fractions were purified by preparative chromatography, microcolumns and 17 secondary metabolites were isolated, 15 of them different. The structural elucidation was performed with the techniques of GC-MS and NMR, allowing the identification of lupenone and lupeol triterpenes, the plant sterol β-sitosterol, the glycosylated flavonoid artubotryside A and a serie of quercetin and kaempferol glycosilated flavonoids.
Description: Se procedió al aislamiento y caracterización de metabolitos secundarios de la especie vegetal Gaiadendron punctatum, utilizada por los indígenas Saraguro en el cantón del mismo nombre, provincia de Loja en Ecuador, para curar distintas afecciones, en especial respiratorias. El procedimiento inició con la recolección de la especie en el cantón Saraguro, para luego secar, pesar y separar las hojas y flores. Inmediatamente se obtuvo el extracto de las hojas mediante maceración con distintos disolventes (hexano, acetato de etilo, metanol, metanol:agua (7:3) y etanol) y el extracto de las flores (hexano, acetato de etilo y metanol). Los extractos de acetato de etilo y etanol de las hojas, y hexano y metanol de las flores se fraccionaron por cromatografía en columna, basados en la riqueza de compuestos dada por cromatografía de capa fina. Las fracciones resultantes fueron purificadas mediante microcolumnas y cromatografía preparativa, aislando 17 metabolitos secundarios, de los cuales 15 son distintos. La elucidación estructural se realizó por medio de RMN y CG-EM, logrando identificar los compuestos: lupeol, lupenona, β-sitosterol, artubotryside A y varios flavonoides glicosidados de la quercetina y kaempferol.
