Abstract: In the present study it was realized the isolation and characterization of native microorganisms of the mining sector Tundayme with capacity to oxidize metallic sulfides; useful for later applications in the biominería. Used medium of cultive 9k to isolate the genre Acidithiobacillus and Leptospirillum with values of pH (6,5, 1.2, 2.4, 1,8) and temperatures (20 and 30°C). Were obtained 48 initial tests of which it selected the 6 better cultives (M1, M3, M5, M7, M9, M11), later determined their oxidating capacities through quantification of the consumption of Fe2+, production of Fe3+ and sulfates. Finally it was managed to obtain gram-negative microorganisms, that present the region 16S rADN with sizes of band approximated to 1500pb; of equal way they presented good capacity to oxidize iron and sulfur; the optimal conditions in culture medium for isolated 1 was pH 1.8 and 30 °C which had volumetric productivity (Qp) of 0.530 g/Lday ferric ion (M7), for isolated 2 were pH 6.5 and 30 °C with volumetric productivity (Qp) was 0.116 g/Lday sulfate (M9) and for isolated 3 were pH 1.2 and 30 °C with volumetric productivity (Qp) of 0.340 g/L ion ferric.
Description: En el presente estudio se realizó el aislamiento y caracterización de microorganismos nativos del sector minero Tundayme con capacidad de oxidar sulfuros metálicos; útiles para posteriores aplicaciones en la Biominería. Se utilizó el medio de cultivo 9k para aislar los géneros Acidithiobacillus y Leptospirillum con valores de pH (6.5, 1.2, 2.4, 1.8) y temperaturas de 20 y 30°C. Se obtuvieron 48 ensayos iniciales de los cuales se seleccionó los 6 mejores cultivos (M1, M3, M5, M7, M9, M11), posterior se determinó sus capacidades oxidativas a través de la cuantificación del consumo de Fe2+, producción de Fe3+ y sulfatos. Finalmente se logró obtener microorganismos gram-negativos que presentan la región 16S rADN con tamaños de banda aproximados a 1500pb; de igual manera presentaron buena capacidad para oxidar hierro y azufre; las condiciones óptimas de crecimiento para el aislado 1 fue de pH1.8 a 30°C el cual presento (Qp)=0.530 g/Ldía de Fe3+ (M7), para el aislado 2 fue pH6.5 a 30°C con (Qp)=0.116 g/Ldía de sulfatos (M9) y para el aislado 3 fue pH1.2 a 30°C con (Qp)=0.340 g/Ldía de Fe3+ (M11).
