Abstract: | Abstract: The physical, chemical and biological analysis of the leaves of essential oil (EO) from the plant Lepecinia paniculata was carried out in the present investigation, the leaves were collected in the canton Saraguro, Loja province. The EO was obtained by steam distillation with a yield of 0.489%, physical analyses performed were specific optical activity, refraction and density index, and the results were -0.3300, 1.4765 and 0.948 g/ml at 20 °C respectively. The chemical composition of oil was evaluated by mass spectrometry coupled gas chromatography (CG-EM), chromatogram for both DB-5ms and HP-Innowax showed the following majority components 1,8-Cineole, -Pinene, -3-Carene, α-Pinene, (E)- Caryophyllene, Guaiol and -Phellandrene. With the analysis to determine the enantiomeric composition, it was possible to identify three pairs of monoterpene enantiomers: (A) (+)-- Pinene, (-)-;-Pinene, (B) (+)--3-Carene, (-)--3-Carene, (C) (+)-Terpinolene, (-)-Terpinolene. Biological activity, the test was carried out against acetylcholinesterase enzymes (AChE) and butyrylcholinesterase (BuChE), results show moderate mean inhibitory concentration (IC50) versus AChE and BuChE, the value of which corresponds to 38.2 ± 2.9 g/mL and 0.14 ± 2.3 g/mL respectively. |
Description: | Resumen: En la presente investigación se determinó la composición química, propiedades físicas y actividad biológica del aceite esencial (AE) de las hojas de la especie Lepechinia paniculata, recolectadas en el cantón Saraguro, provincia de Loja. El AE se obtuvo por destilación por arrastre con vapor de agua, con un rendimiento de 0.489%. Los análisis físicos realizados fueron actividad óptica específica (-0.3300), índice de refracción (1.4765) y densidad (0.948 g/ml). La composición química del aceite se evaluó mediante cromatografía de gases acoplada a espectrometría de masas (CG-EM), el cromatograma tanto para la columna DB5- MS y HP-INNOWAX mostró los siguientes componentes mayoritarios, 1,8-Cineole, -Pinene, -3-Carene, ;-Pinene, (E)-Caryophyllene, Guaiol y β-Phellandrene. Con el análisis de determinación de la composición enantiomérica, se logró identificar tres pares de enantiómeros monoterpenos: (A) (+)--Pinene, (-)-;-Pinene, (B) (+)--3-Carene, (-)-;-3- Carene, (C) (+)-Terpinolene, (-)-Terpinolene. La actividad biológica, ensayo que se realizó frente a las enzimas acetilcolinesterasa (AChE) y butirilcolinesterasa (BuChE, muestran una concentración inhibitoria media (IC50) moderada frente a la AChE y BuChE cuyo valor corresponde a 38.2 ± 2.9 g/mL y 47.4 ± 2.3 g/mL, respectivamente. |
