Abstract: | Abstract: Alzheimer's disease affects 4% of people over the age of 75. It is mainly treated with donepezil, which relieves symptoms but has side effects such as nausea and dizziness due to its synthetic origin, so effective alternatives are needed. Avocado seeds, a by-product that makes up 30% of the weight of the fruit, are typically discarded. To increase shelf life, the seeds were dried at 70°C for 13 hours. Extraction was performed with 31% ethanol using ultrasound, a rotary evaporator and a freeze dryer. Using high performance liquid chromatography, it was found that the ethanolic extract obtained a higher concentration of antioxidant activity, which was fractionated by flash chromatography. They were separated into four different fractions, identifying that fraction D (15-20%) has a higher antioxidant capacity 7789 ± 141 (DPPH) and 213 ± 6 mg GAE/g (TPC), which is characterized mainly by peak 50. Fraction B shows higher anti-acetylcholinesterase capacity in peak 30. This is the first study in which the compounds responsible for antioxidant and anti-acetylcholinesterase activity are fractionated and determined. |
Description: | Resumen: El Alzheimer, afecta al 4 % a personas mayores de 75 años. Principalmente se trata con donepezilo que, aunque alivian los síntomas, tienen efectos adversos nauseas, mareo por su origen sintético, lo que se necesitan alternativas efectivas. Las semillas de aguacate, un subproducto que representa el 30 % del peso de la fruta, que normalmente se desecha. Con la final de incrementar la vida útil se secó durante 13 horas a 70°C. la extracción se realizó usando etanol 31% con ultrasonido, utilizando rotavapor y liofilizador. Mediante cromatografía líquida de alta resolución se determinó que el extracto etanólico obtuvo una mayor concentración de actividad antioxidante, el cual se fraccionó mediante cromatografía flash. Se separaron en cuatro fracciones distintivas, identificando que la fracción D (15-20 %) posee mayor capacidad antioxidante 7789 ± 141 (DPPH) y 213 ± 6 mg GAE/g (TPC) que se caracteriza sobre todo por el pico 50. La fracción B presenta mayor capacidad antiacetilcolinesterasa en el pico 30. Este es el primer estudio en el cual se fracciona y determina los compuestos responsables de actividad antioxidantes y antiacetilcolinesterasa. |
